研究發現STAT1通過啟動甲羥戊酸通路增強YAP1的核定位和轉錄功能。 染色質免疫沉澱實驗驗證STAT1直接結合SREBF1/2基因調控區。 突變KRAS細胞中，STAT1磷酸化缺陷突變體（S727A）無法恢復SREBP表達，而S727磷酸化類比突變體（S727E）同樣無效，提示該位點磷酸化對通路激活的關鍵作用。
YAP1-TEAD4複合體形成正反饋迴路
YAP1在TEAD4協助下直接上調SREBP家族基因表達，形成"STAT1-YAP1-SREBP-甲羥戊酸通路"的正反饋迴圈。 藥理學抑制YAP1-TEAD相互作用（VT104）可破壞該迴路，聯合他汀類藥物（甲羥戊酸通路抑製劑）顯著增強抗腫瘤效果。

耐葯機制突破
揭示突變KRAS癌細胞通過STAT1-YAP1軸產生以下耐藥性：
- 甲羥戊酸通路抑製劑耐葯：雙敲除細胞對cerivastatin敏感性增加3倍
- EGFR靶向治療耐葯：STAT1/YAP1缺失使afatinib IC50降低4.6倍
- 突破傳統KRAS抑製劑耐葯：YAP1啟動可繞過KRAS依賴性
