員表示，他們已確定衰老細胞開始被自身蛋白質廢物"窒息"的關鍵原因，這一發現有助於解釋為何阿爾茨海默病、帕金森病、肌萎縮側索硬化症（ALS）和亨廷頓病等疾病往往在晚年發作。 發表在《衰老細胞》雜誌上的一項研究中，德布拉·托伊伯教授、博士後研究員丹尼爾·斯坦及其合作者報告稱，天然存在的酶SIRT6起到一種細胞交通管制員的作用，控制蛋白質生產並幫助細胞避免有毒物質的積累。
這項研究指向了一個長期困擾科學家的問題。 在遺傳性神經退行性疾病病例中，有缺陷的基因從一開始就可能產生畸形蛋白質。 但大多數病例並非遺傳所致，有害蛋白質團塊出現的原因一直較為模糊。 本-古裡安團隊發現，當SIRT6水準下降——這一過程與衰老有關——細胞對部分蛋白質製造機制失去控制。 它們開始過量、過快地生產，而品質控制系統無法跟上。 結果是錯誤摺疊的蛋白質堆積，細胞難以清除。
這一發現意義重大，因為蛋白質聚集是幾種主要腦疾病的標誌之一。 研究人員長期以來知道這些「殘骸」的存在。 但更難確定的是，為何在許多不攜帶已知遺傳突變的患者中，這些物質會積累。
該研究還提供了一種可能中斷損害的方法。 當研究人員使用美國食品藥品監督管理局（FDA）批准的藥物4PBA來減緩過度的蛋白質生產時，他們能夠減少摺疊壓力並減輕與蛋白質聚集體相關的有害影響。 在秀麗隱桿線蟲（C. elegans）模型中，這種治療方法改善了壽命和活動能力，表明失控的蛋白質生產至少部分驅動了功能衰退。
這些發現並不等同於治癒方法，模型生物中的結果也不總是能完美轉化為人類醫學應用。 儘管如此，該研究為研究人員提供了更清晰的解釋，說明衰老如何加劇神經退行性變——以及嘗試阻止它的更明確目標。
